Rapid test Card for Matrine and Oxymatrine
1.Test Principle
This test strip is based on the principle of competitive inhibition immunochromatography. After extraction, the matrine and oxymatrine in the sample binds to the colloidal gold-labelled specific antibody, which inhibits the binding of the antibody to the antigen on the detection line (T-line) in the test strip, resulting in a change in the colour of the detection line, and a qualitative determination of matrine and oxymatrine in the sample is made by comparing the colour of the detection line with the colour of the control line (C-line).
2.Applications
This kit is used for rapid qualitative analysis of matrine and oxymatrine residues in honey.

6. Sample preparations
(1) To ensure homogeneity of sampling, if honey sample is crystallised, need to warm in water bath at temperature no more than 60°C. The honey samples were melted, stirred and mixed, and then cooled to be room temperature for assay step.
(2) Screw down the cap of the sample bottle on the horizontal table (pay attention to avoid spilling the liquid inside the bottle), and cap of the sample bottle for use.
(3) Use sample spoon (if there is no suitable sample spoon, 3 mL dropper can be used for sampling) to take the honey sample , place it inside the bottle cap to fill the inner tank (about 0.6~0.7g). Then tighten the dissolving bottle cap with the corresponding bottle, shake until the honey sample is completely dissolved, and the liquid in the bottle is the liquid to be assay.
7. Assay process
(1) Read the instructions carefully before experiment. Wait until the test kit and samples to be room temperature.
(2) Take out the card from kit package,Use immediately after marking
(3) Open the yellow cap of the pre-treated vial, squeeze the vial slightly, and add 3 drops of the test solution (about 90-120 μL) into the sample hole.
(4) Start timing when the liquid is flowing, react for 10 min, judge the result according to Part 8.

8.Results

Negative                         Positive                              Invalid
Negative(-) : Color of Line T is deeper than Line C, indicating the matrine and oxymatrine in sample is less than LOD of the kit.
Positive(+) : Line C and Line T are both red, Line C is red, color of line T is weaker than line C or the Line T is colorless, indicating matrine and oxymatrine in sample is higher than LOD of the kit.
Invalid: Line C has no color, which indicates the strips are invalid. In this case, please read the instructions again, and redo the assay with new strip.
9. Notice for operations

1. 1. Please do the assay following the instruction, do not touch the membrane of the strip.
   2. Please use the card and assay solution immediately after open .
   3. Don't use different LOT test card with dissolving Bottle solution.
   4. This strip is used for only once; please do not use it repeatedly.
   5. This kit is only for screening test, positive result should be further confirmed with other method.

10. Specificity
The test shows negative with 500μg/kg of tetracyclines, fluoroquinolones and sulphonamides.
The test shows positive with 10μg/kg of sophocarpine and sophoridine.
11. Storage
2-30°C in cool dark place, do not freeze. The kit will be valid in 12 months. The lot number and expired date are printed on the package.
