Zearalenone Immunoaffinity columns
  1. Principle
The zearalenone immunoaffinity column is based on antigen- antibody-reaction technology, which makes the test highly specific, sensitive, rapid and simple to perform.
The columns contain a gel suspension of monoclonal antibody specific to the toxin of interest. Following extraction of the toxin the sample extract is filtered, diluted and passed slowly through the immunoaffinity column. Any toxin which is present in the sample is retained by the antibody within the gel suspension. The column is washed to remove unbound material and the toxin is then released from the column following elution with solvent. The final elution solution is for assay.
   It can reduce efficiently the sample impurity interference and improve the accuracy of test if it is combined with the HPLC, fluorescence and immunoassay.
  1. Sample
It can test the maize, soybean, peanut, wheat, barley, rice and finished feed.
  1. Required equipment
----Homogenizer
----Vortex mixer

----Measuring cylinder: 10ml, 100ml
----Qualitative filter paper/Centrifuge
----Analytical balance (inductance: 0.01g)

  1. Reagent
----Methanol (AR)
----Sodium chloride (NaCl, AR)
----Deionized water
  1. Reagents preparation
Solution 1: 60% Methanol solution
Mix the deionized water and the methanol in the volume of 4:6. It is used for preparing the sample.
  1. Sample preparations
----Homogenize the sample.
----Take 5.0±0.05g sample into the sample bottle. Add 1.0±0.05g NaCl and 25ml of 60% methanol solution (solution 1) and vortex for 5min or shake for 20min with table concentrator. ----Centrifuge: 5min/3000g at least/room temperature (20-25ºC).(If no centrifuge, it can also stand the solution and take 10ml of the supernatant to filter)
----Take 5ml (similar to 1g sample) of supernatant after centrifuge or filter and add 5ml of deionized water. Mix completely and be for assay.
  1. Assay process
----Take out the columns in the 2-8ºC/35.6-46.4ºF and recover to room temperature (20-25ºC/68-77ºF).
----Connect the columns to the 10ml injector by the joint (picture 1). 
Wash the column with 5ml deionized water for twice.
----Pipette 5ml (0.5g sample) of prepared solution into the injector→→use the injector to control the flow rate of the liquid to 1-2 seconds per drop through the column.
---- Wash the column with 5ml deionized water for twice time →→use the injector to control the flow rate of the liquid to 0.5-1 seconds per drop through the column. Finally drain the liquid in the column.
----Elute with 1ml methanol→→use the injector to control the flow rate of the liquid to 2-3 seconds per drop through the column. Collect all the eluent and be for assay.
Notice:
----Peristaltic pump can also be used to control the flow rate to through the column.
  • ----For a mass of test, using the pump flow operation can reduce the working time and intension.
8. Sample dilute ratio and recovery rate
Sample dilute ratio: 2 time
Recovery rate: ≥85%
9.Storage
Storage at 2-8ºC/35.6-46.4ºF. The period of validity is 12months.
